The first total synthesis of (±)-annosqualine by means of oxidative enamide-phenol coupling: pronounced effect of phenoxide formation on the phenol oxidation mechanism

The first total synthesis of a spiro-isoquinoline alkaloid, (±)-annosqualine, was established by employing an enamide-phenol coupling of a 1-methylene-1,2,3,4-tetrahydroisoquinoline derivative with a hypervalent iodine reagent, where the formation of the phenoxide was recognized to be an essential step for the reaction of the phenolic hydroxyl group with the hypervalent iodine reagent leading to the formation of the desired product.     

Reductive coupling of polyfunctionalized organobismuth and organolead arylating reagents in the synthesis of benzopyran derivatives

Benzopyran derivatives were synthesized in good yields by the reactions of tris[2-(chloromethyl)phenyl]bismuth diacetate and 2-(halomethyl)aryllead triacetates with phenols and naturally occurring 4-hydroxycoumarins in the presence of bases according to a three-step one-pot procedure. A concise and efficient synthesis of salvinal from isoeugenol via a phenoxenium ion intermediate

In this letter, we describe how salvinal can be efficiently synthesized from isoeugenol via a phenoxenium ion intermediate by four steps in 23% over all total yield.     

Microgels and ionic associations in solutions of cellulose diacetate

Solutions of cellulose diacetate (CDA) from two sources (cotton linters and wood pulp Floranier) were analysed in various solvents by size exclusion chromatography (SEC). Without special precautions, the SEC chromatograms presented three peaks — or prehumps — before the main polymer peak. The first prehump which could be eliminated by ultracentrifugation corresponded to microgels whose sugar composition was determined. These microgels were also investigated by electron microscopy, X-ray and electron diffraction analysis. They corresponded mainly to cellulose triacetate (CTA-II) in the case of CDA from cotton linters and a mixture of CTA-II and xylan diacetate (XDA) in the case of CDA from the wood pulp Floranier. The second and third prehumps could be attributed to ionic effects corresponding to the association of remaining sulfate groups on the CDA molecules with residual calcium. It was found that these ionic effects could be eliminated by the addition of LiBr or LiCl to the elution solvents. This led to chromatograms devoid of prehumps.Presented in part at the Cellulose '91 meeting in New Orleans.     

A Novel Method for the Synthesis of Substituted Benzochromenes by Ethylenediamine Diacetate‐Catalyzed Cyclizations of Naphthalenols to α,β‐Unsaturated Aldehydes. Concise Synthesis of the Natural Products Lapachenole,Dihydrolapachenole, and Mollugin

A new synthetic route for biologically interesting benzochromenes was developed starting from naphthalenols and α,β‐unsaturated aldehydes in the presence of ethylenediamine diacetate. This methodology was applied for the total synthesis of the biologically important natural products lapachenole, dihydrolapachenole, and mollugin with a benzochromene moiety.     

16-脱氢孕烯醇酮醋酸酯的环境友好合成法

Instead of environmentally toxic chromium oxidant, singlet oxygen generated photcchemically was used as environmentally friendly and benign oxidizing agent to accomplish the transformation of pseudodiosgenin diacetate to diosone efficiently in a low toxic and less expensive solvent acetone. Accordingly 16-dehydropregnenolone acetate(16-DPA), an important intermediate for preparation of steroidal drugs, was prepared in good yield (75%) when photoreaction was run in acetone/acetic anhydride/pyridine system. The mild reaction condition as well as simple and environmentally friendly process made the method commercially viable and important for production of 16-DPA in industrial scale.     

Chemical and analytical characterization of related organic impurities in drugs

A system is proposed for the classification of related organic impurities in drugs and drug products including among others (separated and non-separated) intermediates, various kinds of by-products, among them products of different side reactions, epimeric/diastereomeric, enantiomeric impurities, impurities in natural products, and finally degradation products. Examples are taken mainly from the author's own experience and from among the named impurities in the European Pharmacopoeia with focus on impurities in hydrocortisone, prednisolone, enalapril maleate, lisinopril, ethynodiol diacetate, pipecuronium bromide, cimetidine, and ethynylsteroids. The methodological aspects of impurity profiling from the detection to the identification/structure elucidation and quantitative determination of impurities are briefly summarized.This paper is Part 23 in the series "Estimation of impurity profiles of drugs and related materials". The extraction,antioxidant and against β-amyloid induced toxicity of polyphenols from Alsophila spinulosa leaves

Alsophila spinulosa is a tree-like fern, and many evidences suggested that plant polyphenols had the potential therapeutic for Alzheimer s disease (AD). Herein, polyphenols (ASP) was isolated from A. spinulosa leaves and its major constituent were isoorientin and vitexin. ASP displayed excellent antioxidant activity and obvious anti-lipid peroxidation capacity in vitro. ASP improved the survival rate of C. elegans under high temperature by enhancing the antioxidant enzymes activities and decreasing the lipid peroxidation level. Moreover, ASP alleviated β-amyloid (Aβ) induced paralysis and reduced Aβ deposition, decreased reactive oxygen species (ROS) accumulation and improved the level of skn-1 mRNA. In addition, ASP decreased the levels of pdk-1 and akt-1 mRNA in P13K/AKT signaling pathway. In conclusion, ASP may be a potential ingredient for the alleviation of AD.     

胆甾-4α-甲基-8-烯-3β，7α-二醇二乙酸酯的结构与性质研究

Lophenol, cholest-4α-methyl-7-en-3β-ol (1), obtained from Dracaena cochinchinensis (Lour.) S. C. Chen, was structurally modified. It was acetylated to protect 3β-hydroxyl group, and then oxidised by selenium dioxide in acetic acid to give cholest-4a-methyl-8-en-3β, Ta-diol diacetate (3). This compound 3 is unstable in chloroform solution or when heated and easily converted to a diene compound, cholest-4a-methyl-7,14-dien-3β-ol acetate (4). The structures of 3 and 4 were elucidated by means of IR, ^1H NMR, ^13C NMR and MS, and the absolute configuration of 3 was established by X-ray crystallography. The property of 3 was also discussed in this paper. Both 3 and 4 are new compounds and were reported for the first time.
